It has since been revealed that the games will feature more than 50 new Pokémon and contain an "innovative communication system" called "Real Play".

"People around the world will be able to simultaneously experience 'Real Play'," reads a statement from Game Freaks president Tsunekazu Ishihara (via Gematsu).

"The game uses an innovative communication system that makes it feel as if you are playing with other players around the world."

Sounds like an interesting step up from the entralink system.
